I have discovered a fatal defect in current management of acute virus infection. For a long time supporting treatment is the main principle for acute virus infection. However limitation of nutrients intake especially protein to prevent replication of virus seems to be the really most important. It is even more important than antiviral medicine treatment. The main structure of a virus is DNA or RNA. DNA and RNA are synthesized from purine and pyrimidine. Purine and pyrimidine are synthesized from amino acids. Amino acids are decomposed from proteins. Obviously protein is the ultimate raw material of virus. From chemical view, if there is no raw material (protein), there is no product (virus). But, what makes us confused is that protein is also the main material for immune cells reproduction and antibodies production. Most people believe that ingestion of protein in acute virus infection can strengthen our immune ability to kill virus. However ingestion of protein in acute stage of virus infection will also make virus replicate massively. Virus antigen is the most potent substance to drive the immune response, so it will induce a very large immune response to kill the massively replicated viruses. Finally it may lead us to suffer so called immune storm. That is, too large immune response damages the tissues of ourselves and finally threatens the patient to death. Prevent ingestion of nutrients to weaken the virus and use the minimized immune power to destroy it is the highest principle of this war. And that is the way in which the immune system always functions.
In fact, evolution has made a perfect arrangement. Our bodies can control virus replication through controlling de novo synthesis of purine and pyrimidine in the liver without interfering with reproduction of immune cells of which de novo synthesis is independent. As a matter of fact, most de novo synthesis of purine and pyrimidine bases occurs in the liver, and the nitrogenous bases and nucleosides are then transported to other tissues by red blood cells. Common cells do not have the ability to do it. They have to rely on the supply of liver. But the immune cells also have the ability to perform de novo synthesis of purine and pyrimidine. Poor appetite when infection is the result of evolution. It is not caused directly by virus. Its purpose is to terminate the ingestion of nutrients, especially protein. At the meanwhile, via decomposing muscle proteins, the serum concentration of amino acids is maintained. Immune cells can obtain them freely for cells reproduction and antibodies production. While in the liver, under the coordination of cytokines(especially Interleukin 1 (IL1)、tumor necrosis factor (TNF) and interleukin 6 (IL6)), sympathetic nerves and stress hormones, the de novo synthesis of purine and pyrimidine is inhibited. So no or very few nucleosides can be synthesized and transported to other cells for the synthesis of DNA and RNA. And virus cannot replicate. However I would like to make a brief summary as following.
First, the digested and absorbed purine and pyrimidine bases and their derivatives are metabolized by epithelial cells of intestine and are expelled through urine. Only about 5 % of purine and pyrimidine contained in the food can enter the circulation and be used. Second, immune system controls the de novo synthesis of purine and pyrimidine bases. It is performed mainly in the liver. Most cells do not have the ability to do it. Third, when in acute virus infection, the appetite is inhibited and intake of protein (amino acids) and other nutrients that are concerned with synthesis of purine and pyrimidine bases are also inhibited. Fourth, continuous decomposition of muscle proteins maintains the serum level of amino acids which can normally be used by immune cells for cells division, antibodies and cytokines production. The immune cells have the ability to de novo synthesize purine and pyrimidine bases. Fifth, increase of energy expenditure forces the amino acids picked up by liver cells to be used to synthesize glucose which is the main energy source in acute stage of virus infection. Fever further enhances this procedure. Sixth, inhibit lysis of fat to fatty acid in adipose tissues and formation of ketone bodies in the liver. So they canít be the energy sources as they are in starvation. Seventh, inhibit LPL (lipoprotein lipase) which can decompose triglyceride of lipoprotein to fatty acid. And triglyceride synthesized from liver canít be energy source, either. Eighth, some of the amino acids picked up by liver cells are used to synthesize triglyceride, which is useless and luxurious at the moment for the LPL is inhibited and nutrients and calories intake are poor. Its purpose is to exhaust amino acids in liver cells. Ninth, some of the amino acids picked up by liver cells are used to synthesize acute phase proteins, which are thought to be helpful to immune reaction. Tenth, no or very few amino acids in the liver cells can be used to synthesize purine and pyrimidine bases. So DNA and RNA canít be synthesized and virus canít replicate.
In the acute stage of virus infection, ingestion of protein must be inhibited (It is against current conception). But there is no need to inhibit protein intake if it is not in the acute stage. Natural foods which contain absolutely no protein are few. Youíd better go to convenience store to buy packaged foods of which ingredients mark protein zero such as fruit jelly, juiceÖand so on. But if an infection goes with high fever, any calorie containing and high purine-pyrimidine foods should also be limited. Theoretically acute virus disease combined treatment with limitation of proteins and related nutrients intake will largely decrease the mortality, contagious ability and severity of influenza, dengue, enterovirus diseases, common cold Ö and so on. It also greatly shortens the total duration of the virus disease.
